-Galactosidase: immunological activity of ribosome-bound, growing polypeptide chains.

Ribosomes carrying nascent chains of beta-galactosidase were prepared by disruption of Escherichia coli in detergent-free buffer of high salt concentration, followed by purification on a discontinuous sucrose gradient. Assay by the method of immune hemolysis inhibition with anti-beta-galactosidase indicated that considerable amounts of antibody were bound by the growing chains. Much of the cros...

Ribosome Function: Governing the Fate of a Nascent Polypeptide

Recent data highlight how eukaryotic ribosomes connect polypeptide synthesis to translational regulation and targeting. Information contained in nascent polypeptides can be transmitted by surprisingly diverse routes.

Nuclear translation visualized by ribosome-bound nascent chain puromycylation

Whether protein translation occurs in the nucleus is contentious. To address this question, we developed the ribopuromycylation method (RPM), which visualizes translation in cells via standard immunofluorescence microscopy. The RPM is based on ribosome-catalyzed puromycylation of nascent chains immobilized on ribosomes by antibiotic chain elongation inhibitors followed by detection of puromycyl...

Atomic force microscopy captures folded ribosome bound nascent chains.

Direct visualization of co-translational folding of nascent polypeptide chains is challenging. Here we present, for the first time, AFM images of large protein constructs based on the membrane binding domain of ankyrin-R, complexed with the ribosome. The characteristic "horse-shoe" shape of ankyrin-R emerging from the ribosome was captured.

Force measurements of the disruption of the nascent polypeptide chain from the ribosome by optical tweezers.

We show that optical tweezers are a valuable tool to study the co-translational folding of a nascent polypeptide chain at the ribosome in real-time. The aim of this study was to demonstrate that a stable and intact population of ribosomes can be tethered to polystyrene beads and that specific hook-ups to the nascent polypeptide chain by dsDNA handles, immobilized on a second bead, can be detect...
